I would like to donate a furniture that is located in the second story of my house. Is there a organization that will pick it up for free AND help me bring it downstairs? Please let me know which.

This is just a guess, but some churches may take donations and help you move it. If not, you could ask them who can.
Volunteers of America will pick it up for you. They’re great.
Salvation Army will arrange a day/time with you and will do all the heavy lifting. It depends on your locale, but many charitable organizations will send a moving truck and a couple of men to get all the items out for you.
You could offer it on Freecycle.com as long as you explain what is needed. Freecycle is people sharing with people, not a charitable organization.
We moved recently, and we found that most charitable organizations (and even private individuals) were quite willing to come pick the stuff up themselves, since you’re giving it away, even if it’s heavy and on the second floor. However, the stuff needs to be in fairly decent condition. They’ll reject it if they don’t think they can resell it.
If you meet their criteria, some jurisdictions will send a work crew of people doing community service to assist you with moving the stuff downstairs.
