Barry Bonds currently holds the MLB home run record with 762.
Most baseball historians would agree that Josh Gibson hit even more than that while playing in the Negro Leagues, however the exact number is unknown since the Negro Leagues did not compile complete statistics or game summaries. Some estimates put the number at 800, even as high as 1,000 career home runs. This research also credits Gibson with a rate of one home-run every 15.9 at bats, which compares favorably with the rates of the top home-run hitters in Major League history.
