Only way I know how (without jailbreaking your phone) is to use the latest version of GarageBand from Apple:

I am able to import any iTunes song, clip the section I want, then export it to iTunes so it can be used as a ringtone. Problem is you have to be on a Mac and you have to pay to have the latest version of GarageBand (it’s not a free upgrade).
@zombo: great reference (my answer too); be aware though that Protected AAC files will not be able to import into iTunes; that is, songs bought in iTunes bear the copy protection that won’t allow the media to be used by any other program.
There are ways around this though (for legal reasons I won’t go into this), but if you dig around you’ll find out on your own.
Take a song into garageband…cut a part to 40 seconds or less export as a AAC. Download Make iPhone Ringtone Drag the AAC you created onto Make iPhone Ringtone and it will import it as a ringtone into itunes….sync your phone….there you go….
if you use a pc, you can buy iphone ringtone maker for $15. I use it, it doesn’t hack the phone in any way, and it works great.

on a pc use the program audacity(free), cut it to how short you want it, export it as mp3. bring it into itunes, select convert to acc. go rename that file to m4r and double click it. boom you have a ringtone! sync away.
